OriAb Antibody Discovery Platform

Our OriAb platform represents one of the largest and most diversified antibody libraries to screen and optimize antibodies for CAR-T programs. OriAb platform comprises 1011 full human naïve phage display antibodies and 1011 synthetic nanobodies. We have mastered know-hows to effectively identify binders with tuned affinities and avidities to target tumor associated antigens and make different characteristics of TAAs to work for our CAR-Ts.

OriAb Antibody Discovery Platform

We have constructed a large scale of 1011 full human naïve phage display library and a 1011 synthetic nanobody library through innovative methods. The synthetic nanobody library, NanoOri_2.0 series in particular, contains both "upright" and "roll" conformations with different length of CDR for screening candidate antibodies against different types of antigens, with sub-libraries especially desiged for CAR-T with low CAR-tonic signaling.

These libraries provide an extraordinary advantage in our mission to discover specific antibodies against various tumor targets. We can efficiently identify a broad spectrum of tumor-specific targets-binding epitopes on a large scale, offering comprehensive candidate sequences. This opens up opportunities to uncover antibodies with superior binding properties, even for challenging targets like GPRC5D. The highly specific antibodies we unearth are proficient at selectively targeting tumor cells while mitigating the risks of CAR-T product self-activation and rapid exhaustion.

In addition, we have designed a unique antibody engineering process, including affinity maturation, humanization, and antibody format modification. This process ensures that we optimize antibodies with the most appropriate affinities for different targets, making them invaluable assets in CAR-T therapies.

CAR-T Platform

Our design incorporates a series of OriCAR signal components, enhancing the cytotoxicity of CAR-T cells against both hematologic and solid tumors through multiple mechanisms. This effectively improves the killing efficacy of CAR-T cells against tumors, reducing the risk of relapse and enhancing tolerance.

OriCAR CAR-T Platform

    OriCAR-017

    ·GPRC5D is a well validated target for Multiple Myeloma as JNJ had launched FiC CD3*GPRC5D BsAb in Aug, 2023.
